Après deux ans d'études de médecine à Yale, Robert Picardo décroche un rôle dans la comédie musicale "Mass", de Leonard Bernstein, et joue même un premier rôle lors de la tournée européenne.
Il poursuit sa carrière à Broadway avec "Gemini" et "Tribute", et remporte un Dramalogue pour l'ensemble de son travail. Ce succès se confirme lorsqu'il se tourne à la télévision, avec une nomination aux Emmy Awards et un prix des téléspectateurs pour ses rôles dans "The Wonder Years" (Les Années Coup de Coeur) et "China Beach".
